I've been playing with PyWinAuto today and having fun automating all sorts GUI tests. I was wondering if it is still state of the art or if there might be something else (also free) which does windows rich client automation better.
pywinauto is great because it's Python.
Perhaps a bit more full featured is AutoIT, which has a COM server that you can automate (from Python using win32com), and some cool tools, like a "Window Info" utility, which will give you the text (title), class, size, status-bar text, and so on for the window currently under the mouse cursor.
There are some cases where pywinauto is a bit harder to use than AutoIt, and seems a little less polished. One example is automating Inno Setup programs. The Inno Setup "setup.exe" program launches a separate application that actually performs the install, and it's a pain to track this down with pywinauto, but AutoIt makes it easy.
I used to do test automation on our projects with AutoIt but switched over to pywinauto 3 months ago and have been very happy with that decision. There are some rough edges, but I've been able to fill them in with my own supplementary test functions. In addition I find that coding tests and support code in Python is much easier and more manageable compared to AutoIt. With Python I have way more powerful options for logging, debugging, documentation, process management and test configuration. For me it was absolutely the right way to go.
I am going the same way, bit by bit and I have to say that python + pywinauto is good stuff!
New pywinauto 0.6.0 has introduced MS UI Automation support under the hood. So that WinForms, WPF, Qt, Store apps etc. could be automated almost the same way as an old native Win32 application.
Moreover it's possible to use mouse and keyboard modules out of a window/control context. These modules work on Linux as well!
New module win32_hooks is inspired by pyHook and similar libraries, but it doesn't require compilation.
Yeah, this post is a kind of ad. But I just wanna say pywinauto is still useful and I believe it will be even more useful in the future. The developers community currently consists of 2 mature developers and 3 talented students and we have long term plans on Linux and Mac OS X.

I have the following data scheme that I want to turn into an application. A regular desktop GUI would probably the best.

The whole thing would be developed on a Linux (Ubuntu, Gnome) machine, and then used on a Windows Vista computer. If Windows is a problem, I might be able to just run it in a Linux VM later.
My programming language of choice would be Python. I have only used Qt for a two hour project, I have pretty much no experience with GUI. Would Qt work easily on Windows and not look to out of place there? Is GTK a way better option, or is that merely preference issue?
From talking on the #python channel, I gathered that storing the data in a SQLite database would be a good option, and accessing this data via the SQLAlchemy ORM would be better than writing SQL statements by hand. The latter is my usual approach, I want to use this project to learn something better.
Can I just design the dialogues in Qt Designer and use them with my Python objects?

If you are set on using Python them I think Qt would be am excellent choice. It's a fantastic framework that looks good and runs on many platforms. You won't be limited to Windows. Qt can be a bit intimidating but you won't be limited in terms of functionality. The Qt designer works well with Python.
SQLAlchemy works on all common desktop platforms, including Windows.
I'm interested in getting started w/ developing Python based applications for a desktop environment and have a few (seemingly simple) questions:
What is the best method for developing GUI applications? I've seen several frameworks but the indexes I've found are a bit convoluted and mix (what seem to be) legacy packages. In your opinion, what is the best approach in this regard?

wxPython is the best GUI framework.
The official docs are the best resource. They helped me quite a bit.
Have you considered Iron Python as an option? It's basically the Python language on top of the .NET Framework. Having been fortunate enough to work with the .NET Framework in the past on desktop-applications, I can attest to its depth of excellent.
wxPython is a phenomenal GUI toolkit for developing native applications. I highly recommend it. Also, if you combine it with py2exe you can create .exe files for running on Windows.
easyGUI and Tkinter are VERY easy libraries that can be used with Python to make GUIs. Further, as stealthdragon has suggested, you can use py2exe to compile your GUIs into EXEs for your PC. There are also other compilers such as pyc.
As Jonathan Sampson suggests, you might consider using IronPython and use it to wield the full power of the .NET framework. The IronPython Cookbook, which among other things shows you how to make a basic Twitter Client.

I'm about to start a new job where the coding practices are heavily centered around TDD and refactoring, and whose primary development language is Python. I come from the Java world, and have been a confident user of Eclipse for a good, long time. When not working in Java, I use emacs.
I'm looking for an IDE for Python that will give me a lot of the capabilities I've grown used to with Eclipse, not only for refactoring but in terms of code completion, project management, SCM integration (currently CVS, but likely to switch to git one of these days) et al.
What IDE should I use?

Have tried many different (Kate, Eclipse, Scite, Vim, Komodo): each one have some glitches, either limited functions, or slow and unresponsive. Final choice after many years: Emacs + ropemacs + flymake. Rope project file open dialog is extremely quick. Rope refactoring and code assist functions are super helpful. Flymake shows syntax mistakes. Emacs is the most configurable editor. I am very happy with this config. I use TextMate for all my Python programming needs. It's not an IDE per se, but it does a lot of stuff that an IDE does (without all the cruft of an IDE). It has syntax highlighting, code folding, integration with various SCMs through the use of additional bundles (I know it supports SVN, Git, Mercurial, Darcs, and probably a few others). It's also quite extensible and customizable (again, through the use of bundles). It also has a basic concept of projects. One place where it doesn't shine, though, is in code completion; some bundles have limited support for code completion, but it's generally not as amazing as that of most language-specific IDEs. Given how awesome TextMate is, though, I don't know sacrificing that. TextMate's definitely made me much more productive.
Pydev for Eclipse, as others have mentioned, is good.
Netbeans has a beta Python plugin that is a little rough around the edges, but could turn into something really cool.
Additionally there is a long list of programming centric text editors for the mac, that may or may not fit your needs.
Textmate - costs money, people love this program, but I haven't used it enough to see what all the fuss is about.
Jedit - Java based text editor, has some nice features, but the startup time isn't great (due to Java).
CarbonEmacs - Decent Emacs port.
AquaEmacs - Better Emacs port.
TextWrangler - Lite, free (as in beer) verision of BBEdit.
BBEdit - The old guard. The defacto editor before Textmate stole its limelight. Expensive.
Smultron - Very nice editor, the UI is similar to Textmate.
Idle - Python's own little editor, has some nice features, but also some major problems. I've personally found it too unstable for my usage.
Sublime Text - This is really sweet text editor that has some surprisingly good Python support.
Pycharm - Another solid full on IDE for Python.
Eclipse with Pydev works best for me on any platform.

I usually use either komodo edit or aquamacs with ropemacs. Although I should warn you, IDE features won't be what you're used to if you're coming from a Java or C# background. I personally find that powerful IDEs get in my way more than they help.
UPDATE: I should also point out that if you have the money Komodo IDE is worth it. It's the paid version of Komodo Edit.

I've used WingIDE and have been very happy. Intellisense is pretty good, some other things are a bit wacky but overall it's a very productive tool
If you are looking for an interactive environment and not needing to code modules, I would suggest IPython. Though this is developed with scientists/statisticians in mind, it will run just as well without any of the scientific packages installed. The features are powerful, with code completion, integrated help, integrated debugging, etc., and it functions as a notebook with Markdown and MathJax integration. By far the best choice for those that need powerful features without wishing to load megabytes of GUI into RAM--since it is browser based, it is used in your always loaded chrome/safari instance. ;-)

Python support on netbeans is surprisingly good, and comes with most of the features you're looking for.
TextMate or Panic's Coda. NetBeans works very well, if you want a full-blown kitchen sink IDE.
"Which editor/IDE for ...?" is a longstanding way to start a "My dog is too prettier than yours!" slapfest. Nowadays most editors from vim upwards can be used, there are multiple good alternatives, and even IDEs that started as C or Java tools work pretty well with Python and other dynamic languages.
That said, having tried a bunch of IDEs (Eclipse, NetBeans, XCode, Komodo, PyCharm, ...), I am a fan of ActiveState's Komodo IDE. I use it on Mac OS X primarily, though I've used it for years on Windows as well. The one license follows you to any platform.
Komodo is well-integrated with popular ActiveState builds of the languages themselves (esp. for Windows), works well with the fabulous (and Pythonic) Mercurial change management system (among others), and has good-to-excellent abilities for core tasks like code editing, syntax coloring, code completion, real-time syntax checking, and visual debugging. It is a little weak when it comes to pre-integrated refactoring and code-check tools (e.g. rope, pylint), but it is extensible and has a good facility for integrating external and custom tools.
Some of the things I like about Komodo go beyond the write-run-debug loop. ActiveState has long supported the development community (e.g. with free language builds, package repositories, a recipes site, ...), since before dynamic languages were the trend. The base Komodo Edit editor is free and open source, an extension of Mozilla's Firefox technologies. And Komodo is multi-lingual. I never end up doing just Python, just Perl, or just whatever. Komodo works with the core language (Python, Perl, Ruby, PHP, JavaScript) alongside supporting languages (XML, XSLT, SQL, X/HTML, CSS), non-dynamic languages (Java, C, etc.), and helpers (Makefiles, INI and config files, shell scripts, custom little languages, etc.) Others can do that too, but Komodo puts them all in once place, ready to go. It's a Swiss Army Knife for dynamic languages. (This is contra PyCharm, e.g., which is great itself, but I'd need like a half-dozen of JetBrains' individual IDEs to cover all the things I do).
Komodo IDE is by no means perfect, and editors/IDEs are the ultimate YMMV choice. But I am regularly delighted to use it, and every year I re-up my support subscription quite happily. Indeed, I just remembered! That's coming up this month. Credit card: Out. I have no commercial connection to ActiveState--just a happy customer.
I've searched on Google for an app like this for a while, and I've found only options with heavy and ugly interfaces.
Then I opened Mac App Store and found CodeRunner. Very nice and clean interface. Support many languages like Python, Lua, Perl, Ruby, Javascript, etc. The price is U$10, but it's worth it!
I've been using an Evaluation copy of Sublime Text. What's good is it doesn't really expire.
It's been good so far and was really easy to get started with.
I may be a little late for this, but I would recommend Aptana Studio 3.x . Its a based on eclipse and has everything ready-to-go for python. It has very good support for DJango, HTML5 and JQuery. For me its a perfect web-development tool. I do HTML5 and Android development too, this way I do not need to keep switching different IDE's. It my all-in-one solution.
Note: you need a good amount of RAM for this to be snazzy !! 4+ GB is awesome !!

You might want to look into Eclim, an Eclipse server that allows you to use Eclipse functionality from within your favorite text editor. For python-related functionality, it uses Rope, PyFlakes, and PyLint under the hood.
I'm planning on using an embedded browser in my pygtk application and I'm debating between gtkmozembed and pywebkitgtk. Is there any compelling difference between the two? Are there any third options that I don't know about?
It should be noted that I won't be using this to access content on the web. I'm mainly using it for UI purposes.
My priorities are:
It needs to be stable.
It needs to be cross-platform.
It should be easy to use.
It should be actively maintained.
It should be extensible.
It should be fast.
gtkmozembed is not available on Windows, although you can use the gecko embedding interface directly. This would require you to write some C++ code.
As far as I know, the gtk webkit port is not available on Windows yet, and still appears to be undergoing a lot of change.
For an example of a cross-platform gecko embedding solution, check out Miro.
Miro is python, and they've written just a couple of C++ classes to embed gecko on Windows, while using gtkmozembed on linux.

Simple question:
What Python GUI API's are out there and what are the advantages of any given API?
I'm not looking for a religious war here, I'm just wanting to get a good handle on all that is out there in terms of Python GUI APIs.
Here's a good list.
I've used Tkinter and wxPython. Tkinter is quite basic, and doesn't use native widgets. This means that Tkinter applications will look the same on any platform – this might sound appealing, but in practice, it means they look ugly on any platform :-/ Nevertheless, it's pretty easy to use. I found Thinking in Tkinter very helpful when I was learning, because I'd never done any GUI programming before. If things like frames and layout algorithms and buttons and bindings are familiar to you, though, you can skip that step.
You can augment Tkinter with Tix (but be warned, Tix doesn't play well with py2exe). Also check out Python Megawidgets, which builds some more advanced controls using the Tkinter basics.
Finally, Tkinter plays nice with the shell: you can start the interpreter, do things like 'import tkinter' 'tk = tkinter.Tk()' etc. and build your GUI interactively (and it will be responsive). (I think this doesn't work if you use IDLE, though)
wxPython is much better looking, and ships with a much greater range of controls. It's cross-platform (though it seems a bit finicky on my Mac) and uses native controls on each platform. It's a bit confusing, though. It also ships with a demo application that shows off most of its features, and provides a test-bed for you to experiment. Some specific thoughts on wxPython:
There are three (?) different ways to lay widgets out. Ignore two of them; just use Sizers. And even then, you can do just about any layout using only BoxSizer and GridBagSizer.
All wx widgets have IDs. You don't need to care what the IDs are, but in the old days (I think) you did need to know, so some old code will be littered with explicit ID assignments. And most demo code will have -1 everywhere as the ID parameter (despite the fact that the methods all have ID as a keyword parameter that defaults to -1 anyway).
Make sure you get the standard wxWidgets docs as well as the wxPython Demo – you need them both.
If you want to use wxPython with py2exe and you want it to look good on Windows XP, you need a bit of trickery in your setup.py. See here
PyQt is excellent if you have experience or interest in Qt.

Most python GUI APIs will be wrappers around the most common c/c++ GUI APIs. You've got a python wrapper for gtk, a python wrapper for qt, a python wrapper for .NET, etc etc.
So really it depends on what your needs are. If you are looking for the easiest way to draw native-looking widgets on Linux, Mac, and Windows, then go with wxPython (python wrapper for WX Widgets). If cross-platform isn't one of your needs though, other libraries might be more useful.
Instead of posting a list of your options I will give my humble opinion:
I am in love with wxPython.
I have used Qt in C++ and Tk way back in the Tcl days but what really makes me like wxPython is the demo that you get with it. In the demo you can browse through all the different widgets frames etc that are part of the framework see the source code and actually see how it looks while it is running.
I had some problems getting the Linux version build and installed but now that I have it available I use it all the time. I have used wxPython for small data analysis applications and I have written several internal tools related to comparing test results, merging source code etc.
I found this link a long time a go: http://www.awaretek.com/toolkits.html. It suggests a tookit based on your criteria. For me it suggests wxPython all the time. Anyway it gives you a bunch of scores on the various toolkits. What is right for me may not be right for you. But it gives you how all the tookits scored according to your criteria, so if you don't like the top toolkit for some reason you can see which ones are closest to your criteria.
QT/GTK/WxWidgets (formerly wxWindows) seem to be among the most mature cross platform GUI toolkits. The only issue is that none is installed with the default installation of Python, so you may have to compile the libraries. If you want something with no installation required that just runs, then go with TKInter because as has been mentioned it is installed by default with Python.
Anyway my criteria were 9 on Ease of Use, 10 on maturity of documentation/widgets, 10 on installed base, 5 on gui code generators, 10 on native look and feel for both windows/linux and 1 and 5 for the last two, I'm not big into Mac OSX (even with a 10 here it suggests wxpython).
PythonCard is really easy to use. That's what I would recommend.

EasyGUI is different from other GUIs in that EasyGUI is NOT event-driven. It allows you to program in a traditional linear fashion, and to put up dialogs for simple input and output when you need to. If you have not yet learned the event-driven paradigm for GUI programming, EasyGUI will allow you to be productive with very basic tasks immediately. Later, if you wish to make the transition to an event-driven GUI paradigm, you can do so with a more powerful GUI package such as anygui, PythonCard, Tkinter, wxPython, etc.

WX has issues on the Mac.
I had a look here, as I want to get an event driven GUI API to do some stuff in Python. I have wx installed on my mac as part of MatPlotLib, but it does not work properly. It wont take in put from the keyboard. I have installed this three times on three different Mac operating systems, and though it worked the first time, the other two times I had this issue.
This version I am using with Enthought's distribution, so no installation was necessary. When I have installed it separately, there were so many dependent installations, that it was a trial to install.
From what I have read here, I will give Tkinter a go, as this needs to be simple and cross platform, but I thought I would just share the above with you. I like the Mac OS for a number of different reasons, but python tools install far easier on Windows (and probably other Linux). I just thought I would give a Mac perspective here.
I like wxPython or Tk.
Tk comes with the standard Python distribution so you don't need install anything else.
wxPython (wxWigets) seems much more powerful and looks a lot nicer. It also works well cross-platform (though not perfectly because it uses different underlying graphic API's on diff system types)
I prefer PyGTK, because I am a GNOME guy. Using PyGTK feels very pythonic to me. The code organization feels consistent, the documentation is clean and thorough, and it's a very easy toolkit to get used to (except for maybe Treeviews).
An easy to use GUI creator for Python doesn't exist. That's amazing really considering small scripting languages like AutoIt and AutoHotkey have great and very simple to use GUI makers. Come on, Python followers, can't you do better?
I've been working with wxPython for a few years now and I like it quite a bit. The best thing about wxPython is that the UI feels native on the different platforms it runs on (excellent on Windows and Linux though not as good on OS/X).
The API lacks some consistency, but you quickly get used to it.
You can check out Testuff (shameless plug, as it's my own product) to get a feeling of what can be done with wxPython (although I must say, with quite a bit of effort).
wxPython, and I'm assuming PyGTK also, can use wxGlade to help you design most UIs you will create. That is a big plus. You don't have to learn how to hand-code the GUI until you're ready. I made several GUI programs straight from wxGlade before I was comfortable enough in how wxPython worked to take a shot at hand-coding.
PyQt has a similar graphic layout device but I've never had good luck getting PyQt to compile correctly. There was also a lack of tutorials and documentation that showed how to create the final Python code; many of the documents I found referred to the C++ version of Qt.
Tkinter is good for quick and dirty programs but, realistically, if you use wxGlade it may be faster to make the program with wxPython. At a minimum, you can use wxGlade to show a visual representation of the program to a client rather than take the time to hand-code the "dummy" program.
There are python-specific gui-api such as kivy (successor or pymt), pygui (based on pyrex), pyui and nufox, which do not compare with the more robust toolkits like wxpython, pyqt, pygtk and tkinter.
They are just extra optional tools.
The only thing unique about them is these are python-specific api, just like there are prima (perl-specific api) and shoes (ruby-specific api). It helps us to understand that when tk is tcl-based port of api (and others are c and c++ based), then these api are specifically done for the respective three scripting languages.
Out of these, kivy is the most robust, whereas pygui's coding is mentioned to be very python-like, pyui is least robust but worth trying and all of these should be portable wherever python or python-based application goes.
Then there is jpype which is a toolkit usable with jython and pydev, and which is actually java's japi customized under python/jython-interface.
